Acetylcholinesterase-staining solution preparation

Solutions Volume or weight
0.1 M acetate buffer pH 6.0
 0.2 M sodium acetate (1.64 g in 100 ml) 192 ml
 Distilled water 200 ml
 0.2 M acetic acid* 8 ml
Incubating solution
 1 Acetyl thiocholine iodide 10 mg
 2 0.1 M acetate buffer (pH 6.0) 6.5 ml
 3 0.1 M sodium citrate 0.5 ml
 4 30 mM copper sulphate 1.0 ml
 5 Distilled water 1.0 ml
 6 5 mM potassium ferricyanide 1.0 ml
  • * 0.2 M acetic acid was added until the correct pH was reached (pH 6.0).

  • The components were added in the order enumerated and mixed well at each stage.
