A multiplex PCR-based protocol for identification and quantification of Vibrio harveyi-related species
2015, AquacultureCitation Excerpt :The multiplex PCRs produced amplicons that were easily visible in agarose gels for template quantities ranging from 300 ng to 300 pg DNA, but products were not visible when the template quantity was reduced to 30 pg DNA (data not shown). The 100-fold reduction in sensitivity of the multiplex PCR compared to the corresponding monoplex reactions is consistent with previous studies (Jackson et al., 1996; Tsai et al., 1994). The sensitivity and reliability of the MPN–multiplex PCR protocol was tested by seeding samples with live cells of the four species V. campbellii, V. harveyi, V. owensii and V. rotiferianus in equal proportions.
