While Luminex was not the first to use beads in flow cytometry applications, as the literature describes previous use as calibrators, in single plex assays and some multiplexed assays based on size differentiation, the data quality and reliability did not compete with established platforms (ELISA) or emerging technologies (planar arrays) [6–9]. During the design phase, Don Chandler realized a smaller bead would not hold adequate dye volumes to generate hundreds of discrete spectral addresses, whereas a larger bead would have reduced uniformity of the carboxylate loading on the bead surface.
